The candidate will use prostate-tumor bearing mice to evaluate the ability of antigen-specific T cells, isolated and expanded from peripheral tissues, to be used to treat these tumors. The candidate will use T-cell receptor sequencing to characterize antigen-specific CD8 T cells, and use this information to quantify antigen-specific T cells in peripheral blood samples and tumor samples from experimental murine models and from human clinical trials. The candidate will evaluate chemically modified peptides for their ability to elicit peptide-specific immune responses using murine and human blood samples, and evaluate these modified peptides as anti-tumor vaccines using laboratory mice bearing prostate tumors. All of these studies will involve handling laboratory animals, and will require experience with flow cytometry, cell sorting, genetic sequencing, and immunological methods of analysis. Laboratory mouse methods (vaccination, intravenous delivery, necropsy), flow cytometry and methods of analysis, T cell receptor sequencing, ELISPOT, ELISA, and cell culture.
