General Summary:
Vertex Cell and Genetic Therapy (VCGT) is seeking a highly creative and technologically curious Associate Scientist for Cell & Gene Analytical Technology (CGAT) Team to make valuable contribution in automated cell-based potency assay development, implementation and testing. This role will assist in the development, implementation and execution of characterization and release assays for identity, purity, strength, and potency for Vertex preclinical, clinical, and next generation cell and gene therapy programs.
The successful candidate has a proven track record of flow cytometry related techniques and understanding of cell and gene therapy, analytical assay development and assay automation to support evaluation and implementation of novel high throughput (HTP) and automated analytical technologies and platforms for Cell Purity and Phenotype.
Key Duties and Responsibilities:
- Assist in flow cytometry assay development for hematopoietic stem cells, human embryonic stem cells and other related cell types.
- Provide routine analytical testing for in-process, stability, and drug substance samples.
- Assist in transfer method from Cell Therapy Analytical Team for assay automation to QC.
- Assist in building the assay application software for end-to-end assay automation.
- Perform either HTP or automated or both test method qualification to establish specificity, sensitivity, linear range, precision, and accuracy of test methods.
- Perform troubleshooting under guidance of SME and maintain trend of the assay performance.
- Handle analytical reagents inventory including reference standard materials as per regulatory standards.
- Maintain excellent written documentation of completed experiment (ELN, LIMS) and maintain knowledge share systems (MS-Teams and share points).
Knowledge and Skills:
- Hands on experience with flow cytometry technique and flow data analysis.
- Hands on experience with mammalian cell culture using aseptic techniques.
- Some knowledge of sample management and ELN/LIMS systems.
- Familiarity with Flow Cytometers (such as FACSLyric, Attune NxT) and their software (such as FlowJo and FACSuite) and/or high content cell imagers (such as Celigo, Arrayscan XTI, CellVista, etc.).
- Basic understanding of robotic liquid handers (Hamilton, TECAN, FACSDuet or other) and their operating software (VENUS, FACSLINK Fluent Gx or other) for automated sample preparation.
- Working knowledge of applicable software such as Python, Excel-macros, Spotfire, etc.
- Basic knowledge of technical document writing including technical reports, qualification protocols and qualification reports.
- Excellent communication and presentation skills, work effectively as a member of a multidisciplinary team, detail oriented, multitasking, goal, and timeline oriented.
Education and Experience:
- Bachelor's Degree with 0-2 years of relevant experience.
